Materials and Methods Materials and methods were detailed in chapter III. Presence or absence of primary xylem tracheary elements was determined in 900 juice vesicle stalks from the middle 1 cm of 30 mature grapefruit by staining fresh cut stalks with 27. phloroglucinol in 957. et...hanol and concentrated HC1. Stalks in which xylem elements were found were washed with deionized water, stained with toluidine blue and fresh mounted for photographs. Absence of phloem cells was verified by observation of preserved fruit tissue.
Observations of plasmodesmata in cells of juice vesicle stalks were taken from ultrathin sections (approximately 60-70 nm) of stage II fruit (4-5 months) cut with a diamond knife and mounted on 300 mesh copper 37 grids. These sections were stained with uranyl acetate and lead citrate followed by carbon coating for stabilization.
